Research
The aim of my master’s thesis is to utilize TOF-SIMS (time-of-flight secondary ion mass spectrometry) to study lipid composition and turnover in human NPCs and mature oligodendrocytes and how the lipid turnover and composition can be normalized in SGS (Schinzel-Giedion syndrome) using ASO (Antisense oligonucleotides) treatment. Furthermore, the lipid composition and turnover of SGS and healthy mouse oligodendrocytes will be studied with and without ASO treatment since the mouse model is currently used as an in vivo model of SGS. 

The key skills I can acquire through my master’s thesis are cell culturing, neuronal cell  differentiation and maturation (NPC into mature oligodendrocytes), isolation and culturing of mouse healthy and SGS oligodendrocytes, treatment of cells using ASOs and control ASO, study lipid turnover using incubation with isotopically 13C-labeled fatty acids, studying plasma membrane lipid distribution using ToF-SIMS imaging and data analysis using principal component analysis. Furthermore, development of general laboratory techniques such as optimization of genotyping, qPCR, immunostaining procedures, plunge freezing, and fluorescence microscopy imaging are also involved in my project.
